Description

IL13RA1 (Interleukin-13 receptor subunit alpha-1), also known as CD213a1 and CT19 (cancer/testis antigen 19), is a subunit of the IL13 receptor complex. The human IL13RA1 is a single-pass type I transmembrane protein belonging to the type I cytokine receptor family. The extracellular domain (ECD) of IL13RA1 contains 3 fibronectin type III domains, D1, D2, and D3, of which the last two comprise the cytokine receptor homology modules (CRHs). IL13RA1 binds IL13 with a low affinity by itself and forms a functional receptor for IL13 together with IL4RA, a subunit shared by IL13 and IL4 receptors. The biological effects of IL13 and IL4 are closely related in part due to a shared receptor system.  IL-13 binds two types of receptor: (1) a heterodimer of the IL13 RA1 and the IL4RA, transducing the IL13-dependent proliferative signal; and (2) the IL13RA2, acting as a nonsignaling "decoy" receptor.  IL13RA1 also serves as an alternate accessory protein to the common cytokine receptor gamma chain for IL4 signaling, but cannot replace the function of IL2RG in enhancing IL2 binding activity. IL13RA1 has been shown to bind tyrosine kinase TYK2, and thus may mediate the signaling processes that lead to the activation of JAK1, STAT3 and STAT6 induced by IL13 and IL4.
