PSMA (prostate-specific membrane antigen), also known as FOLH1 (folate hydrolase 1), NAALAD1 (N-acetylated-α-linked acidic dipeptidase I), GCPII (glutamate  carboxypeptidase II), FGCP (folypoly¬γ¬glutamate carboxypeptidase ), and GIG27 (cell growth-inhibiting gene 27 protein), is a tumor marker in prostate cancer.  PSMA is a single pass type II transmembrane zinc metallopeptidase that belongs to the peptidase M28 family and M28B subfamily. PSMA is most highly expressed in the nervous system, prostate, kidney, and small intestine. PSMA has folate hydrolase and N-acetylated-α-linked-acidic dipeptidase (NAALADase) activity with a preference for tri-alpha-glutamate peptides. In the intestine, PSMA hydrolyzes folylpoly¬γ-glutamates, facilitating the uptake of folate. Genetic variation in PSMA may be associated with low folate levels and consequent hyperhomocysteinemia. In the brain, PSMA modulates excitatory neurotransmission through the hydrolysis of the neuropeptide, N-aceylaspartylglutamate (NAAG), thereby releasing glutamate, another neurotransmitter. Inhibition of brain PSMA activity is considered to be a promising approach for the treatment of neurological disorders associated with glutamate excitotoxicity, such as stroke, chronic pain, and amyotrophic lateral sclerosis. PSMA also exhibits a dipeptidyl-peptidase IV (DPPIV) type activity. PSMA is involved in tumor progression In the prostate and used as an effective diagnostic and prognostic indicator of prostate cancer.

Construct Details: Expressed as a single-chain Fv (scFv) from anti-PSMA VH and VL region fused with a scFv of anti-CD3e VH and VL region with a His-tag at the C-terminus. Exists as a monomer under non-reducing and reducing condition.

 

Source: Mammalian cells stably expressing anti-PSMA:anti-CD3e bispecific antibody (in a scFv-scFv’ format) and growing in chemical-defined media with no animal component or antibiotics.

 

M.W.: Calculated molecular mass (kDa): 57.0. Estimated by SDS-PAGE under non-reducing condition(kDa): 55-60. Calculated extinction coefficients (M-1 cm-1, at 280nm):  121295.

 

Purity: >95% judged by SDS-PAGE under reducing condition (see the gel image above, labeled as DTT "+")

 

Formulation: Supplied at 0.5 mg/ml in sterile PBS pH7.4 (concentration determined by UV spectrometry and verified by SDS-PAGE and Coomassie blue staining).

Biological Activity: Recognizes PSMA on tumor cells and CD3e in the context of TCR complex on T cells.  Potently promotes T cell-mediated anti-tumor activity in vitro and in vivo after engaging with tumor cell surface antigen PSMA.

 

 

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
