PSMA (prostate-specific membrane antigen), also known as FOLH1 (folate hydrolase 1), NAALAD1 (N-acetylated-α-linked acidic dipeptidase I), GCPII (glutamate  carboxypeptidase II), FGCP (folypoly¬γ¬glutamate carboxypeptidase ), and GIG27 (cell growth-inhibiting gene 27 protein), is a tumor marker in prostate cancer.  PSMA is a single pass type II transmembrane zinc metallopeptidase that belongs to the peptidase M28 family and M28B subfamily. PSMA is most highly expressed in the nervous system, prostate, kidney, and small intestine. PSMA has folate hydrolase and N-acetylated-α-linked-acidic dipeptidase (NAALADase) activity with a preference for tri-alpha-glutamate peptides. In the intestine, PSMA hydrolyzes folylpoly¬γ-glutamates, facilitating the uptake of folate. Genetic variation in PSMA may be associated with low folate levels and consequent hyperhomocysteinemia. In the brain, PSMA modulates excitatory neurotransmission through the hydrolysis of the neuropeptide, N-aceylaspartylglutamate (NAAG), thereby releasing glutamate, another neurotransmitter. Inhibition of brain PSMA activity is considered to be a promising approach for the treatment of neurological disorders associated with glutamate excitotoxicity, such as stroke, chronic pain, and amyotrophic lateral sclerosis. PSMA also exhibits a dipeptidyl-peptidase IV (DPPIV) type activity. PSMA is involved in tumor progression In the prostate and used as an effective diagnostic and prognostic indicator of prostate cancer.

Construct Details: Expressed as the combination of a heavy chain (HC) containing VH from anti-human PSMA mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-human PSMA mAb and CL of human kappa light chain.  Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition (see the gel image above, labeled as DTT "-").

 

Source:  Mamamlian cells stably expressing anti-human PSMA human heavy chain and light chain and growing in chemical-defined media with no animal components or antibiotics 

 

M.W.: Calculated molecular mass (kDa): 146 (49 kDa for a single HC, 24 kDa for a single LC); Estimated by SDS-PAGE under reducing condition(kDa): 50 kDa + 25 kDa.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
