Description

CSF1 (colony stimulating factor 1), also known as MCSF (macrophage colony stimulating factor) is a single-pass type I transmembrane or secreted protein. The active form of CSF1 is a disulfide-linked homodimer, produced by proteolytic cleavage of membrane-bound precursors.  As a cytokine, CSF1 controls the production, differentiation, and function of macrophages. CSF1 primes and enhances macrophage killing of tumor cells and microorganisms, regulates the release of cytokines and other inflammatory modulators from macrophages, and stimulates pinocytosis. CSF1 induces the release of proinflammatory chemokines, and thereby plays an important role in innate immunity and in inflammatory processes. It also plays an important role in the regulation of osteoclast proliferation and differentiation, the regulation of bone resorption, and is required for normal bone development. CSF1 is required for male and female fertility. CSF1 induces reorganization of the actin cytoskeleton, regulates formation of membrane ruffles, cell adhesion and cell migration. Aberrant expression of CSF1 or its receptor CSF1R can promote cancer cell proliferation, invasion and formation of metastases. Overexpression of CSF1 or CSF1R is observed in many types of cancers including breast, ovarian, prostate, and endometrial cancers.  Aberrant expression of CSF1 or CSF1R is implicated in inflammatory conditions, such as rheumatoid arthritis (RA), glomerulonephritis, atherosclerosis, and allograft rejection.
