Description

ST14 (suppressor of tumorigenicity 14 protein) is an epithelial-derived, 1 pass type II transmembrane serine protease belonging to the peptidase S1 family. ST14 contains 2 CUB, 4 LDL-receptor class A, 1 peptidase S1 and 1 SEA domains in the extracellular region. ST14 may form a complex with the Kunitz-type serine protease inhibitor, HAI-1, and is found to be activated by sphingosine 1-phosphate. This protease has been shown to cleave and activate hepatocyte growth factor/scattering factor, and urokinase plasminogen activator, which suggest the function of this protease as an epithelial membrane activator for other proteases and latent growth factors. ST14 degrades extracellular matrix and exhibits trypsin-like activity as defined by cleavage of synthetic substrates with Arg or Lys as the P1 site. ST14 mat be involved in the terminal differentiation of keratinocytes through prostasin (PRSS8) activation and filaggrin (FLG) processing.  The expression of ST14 has been associated with breast, colon, prostate, and ovarian tumors, which implicates its role in cancer invasion, and metastasis.  A mutation in ST14 is associated with autosomal recessive ichthyosis with hypotrichosis.

Construct Details: Full length human ST14 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
