TPBG (trophoblast glycoprotein), also known as oncofetal antigen 5T4 and WAIF1 (wnt-activated inhibitory factor 1), is a single-pass type I transmembrane glycoprotein of the LRR (leucine-rich repeat) protein family.  TPBG is an oncofetal antigen that is specific to trophoblast cells except for amniotic epithelium. TPBG is expressed by all types of trophoblasts as early as 9 weeks of development.  In adults its expression is limited to a few epithelial cell types but highly abundant in many types of  tumor cells. TPBG’s expression is associated with poor clinical outcome in numerous cancers.  TPBG contains 1 LRRNT (LRR N-terminal cap), 7 LRR (LRR1-7), and 1 LRRCT (LRR C-terminal cap) domains in the extracellular region. TPBG may function as an inhibitor of Wnt/beta-catenin signaling by indirectly interacting with LRP6 and blocking Wnt3a-dependent LRP6 internalization.  The C-terminal part of LRRNT and LRR1 are essential for the inhibition of the Wnt signaling pathway.  TPBG may also be involved in cell adhesion and motility.

Construct Details: The recombinant human TPBG-Fc fusion is expressed as a 561 amino acid protein consisting of Ser32 - Ser355 region of TPBG and a C-terminal Fc from human IgG1, which exists as a dimer under non-reducing condition.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
