LILRB2 (Leukocyte immunoglobulin like receptor B2), also known as CD85d, LIR2 (Leukocyte immunoglobulin-like receptor 2), ILT4 (Ig-like transcript 4), and MIR10 (myeloid inhibitory receptor 10), is a member of the leukocyte immunoglobulin (Ig)-like receptor (LIR) family of the Ig superfamily (IgSF).  It belongs to the subfamily B class of LIR receptors (LILRBs), which are single-pass type I trnasmembrane proteins with 2 or 4 extracellular Ig-like domains, a transmembrane domain (TM), and 2-4 cytoplasmic immunoreceptor tyrosine-based inhibitory motifs (ITIMs). LILRB2 has 4 Ig-like C2 type domains and 3 TIIMs. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases to modulate cellular responses.  LILRB2 is expressed predominantly on B-cells and monocytes, and at lower levels on dendritic cells. It is also detected at low levelson natural killer (NK) cells.  Like LILRB1, LILRB2 binds to MHC class I molecules on antigen-presenting cells (APCs) and transduces a negative signal that inhibits stimulation of an immune response. LILRB2 may control inflammatory responses and cytotoxicity to help focus the immune response and limit autoreactivity. LILRB2 recognizes a broad spectrum of HLA-A, HLA-B, HLA-C and HLA-G alleles. Engagement of LILRB2 present on natural killer cells or T-cells by class I MHC molecules protects the target cells from lysis. LILRB2 also binds FCGR1A.

Construct Details: Full length human LILRB2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

 

Vector Type: pLTC (lentiviral expression vector containing a heterologous CMV promoter and with or without a marker, see the vector map above)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
