GFP (green fluorescent protein) was discovered in Aequorea victoria (Jellyfish).  GFP transduces the blue chemiluminescence of aequorin into green fluorescent light upon receiving energy from the Ca2+-activated photoprotein aequorin. Its chromophore is formed by autocatalytic backbone condensation between Ser-65 and Gly-67, and oxidation of Tyr-66 to didehydrotyrosine. GFP has been engineered to produce a large number of differently colored variants for making chimeric proteins as a fluorescent protein tag. GFP proteins can be expressed in most known cell types as a noninvasive marker in living cells and organisms. Therefore GFP proteins enable a wide range of applications where they can act as a cell lineage tracer, reporter of gene expression, as a measure of protein-protein interactions, or as a molecular biosensor and thermometer.

Construct Details: GFP (encoding UniProt #P42212) is cloned into the Lentiviral expression vector pLTC with a CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It was co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of antibiotic stable-selection markers are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
