Description

ABCG2 (ATP-binding cassette sub-family G member 2) or CD338, is a six-pass type II transmembrane glycoprotein beloning to the ABC transporter superfamily. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White/ABCG). ABCG2 is a member of the White/ABCG subfamily. ABCG2 functions as a xenobiotic transporter which may play a major role in multi-drug resistance. It is mplicated in the efflux of numerous drugs and xenobiotics: mitoxantrone, the photosensitizer pheophorbide, camptothecin, methotrexate, azidothymidine (AZT), and the anthracyclines daunorubicin and doxorubicin.  ABCG2 exists as a monomer under reducing conditions, but the minimal functional units is a homodimer; disulfide-linked. The major oligomeric form in plasma membranes is a homotetramer with possibility of higher order oligomerization up to homododecamers.  The extracellular loop 3 (ECL3) is involved in binding porphyrins and transfer them to other carriers, probably albumin.  ABCG2 is also a high-capacity urate exporter functioning in both renal and extrarenal urate excretion. ABCG2 plays a role in porphyrin homeostasis as it is able to mediates the export of protoporhyrin IX (PPIX) and heme. Xenobiotic transporter that may play an important role in the exclusion of xenobiotics from the brain.

Construct Details: Full length human ABCG2/CD338 is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
