Description

DDR1 (epithelial discoidin domain receptor 1) receptor tyrosine kinase (RTK), is a single-pass type I transmembrane glycoprotein that belongs to the insulin receptor (InsR)subfamily of the tyrosine protein kinase family. DDR1comprises a 397-aa extracellular domain (ECD), a transmembrane region (TM), and a 475-aa intracellular kinase domain.  DDR1 may play a key role in the communication of cells with their microenvironment and be involved in the regulation of cell growth, differentiation and metabolism. Like other RTKs, DDR2 transduces signals across the membrane has been shown to be ligand induced receptor oligomerization and subsequent intracellular phosphorylation. This autophosphorylation leads to phosphorylation of cytosolic targets as well as association with other molecules, which are involved in pleiotropic effects of signal transduction.  DDR1shows homology to Dictyostelium discoideum protein discoidin I in their extracellular domain, and that are activated by various types of fibrillar collagen. Expression of this protein is restricted to epithelial cells, particularly in the kidney, lung, gastrointestinal tract, and brain. In addition, it has been shown to be significantly overexpressed in several human tumors. DDR1 regulates cell attachment to the extracellular matrix, remodeling of the extracellular matrix, cell migration, differentiation, survival and cell proliferation. DDR1 is required for normal blastocyst implantation during pregnancy, for normal mammary gland differentiation and normal lactation. It is also required for normal ear morphology and normal hearing.  DDR1 promotes smooth muscle cell migration, and thereby contributes to arterial wound healing. DDR1 also plays a role in tumor cell invasion.

Construct Details: Full length human DDR1 (Uniprot# Q08345-2) gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
