Description

ACKR3 (atypical chemokine receptor 3) is a 7-transmembrane protein of the ACKR subfamily of the G protein-coupled receptor 1 (GPCR1) family.  ACKR controls chemokine levels and localization via high-affinity chemokine binding that is uncoupled from classic ligand-driven signal transduction cascades, resulting instead in chemokine sequestration, degradation, or transcytosis. ACKR is also known as interceptor (internalizing receptor) or chemokine-scavenging receptor or chemokine decoy receptor. ACKR3 acts as a receptor for chemokines CXCL11 and CXCL12/SDF1. Chemokine binding does not activate G-protein-mediated signal transduction but instead induces beta-arrestin recruitment, leading to ligand internalization and activation of MAPK signaling pathway. ACKR3 is required for regulation of CXCR4 protein levels in migrating interneurons, thereby adapting their chemokine responsiveness. In glioma cells, ACKR3 transduces signals via MEK/ERK pathway, mediating resistance to apoptosis. ACKR3 promotes cell growth and survival but is not involved in cell migration, adhesion or proliferation of normal hematopoietic progenitors. ACKR3 plays a regulatory role in CXCR4-mediated activation of cell surface integrins by CXCL12. ACKR3 is required for heart valve development. ACKR3 acts as a co-receptor with CXCR4 for a restricted number of HIV isolates. Translocations involving the ACKR3 gene and HMGA2 on chromosome 12 have been observed in lipomas.

Construct Details: Full length human ACKR3 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
