Description

BCMA (B cell maturation antigen), is a single-pass, type III transmembrane glycoprotein in the TNF receptor superfamily (TNFRSF17). BCMA contains a short extracellular domain (ECD) of 84 amino acid (aa), containing one TNFR cysteine-rich repeat. Within the TNFRSF, it shares the highest homology with TACI (TNFRSF13B). Both BCMA and TACI have been shown to bind to APRIL (TNFSF13) and BAFF (TNFSF13B), members of the TNF ligand superfamily. BCMA is expressed in immune organs and mature B cells, but not in T-cells or monocytes. BCMA is located on the cell surface and also found in a perinulear Golgi-like structure.  The binding of BCMA to APRIL or BAFF has been shown to stimulate IgM production in peripheral blood B cells and increase the survival of B cells, suggesting that BCMA may play an important role in B cell development,  function and regulation. BCMA lacks a death domain (DD) in the cystoplasmic region.  BCMA interacts with various TNFR-associated factors (TRAFs), including TRAF1, TRAF2 and TRAF3, to transduce signals for B cell survival and proliferation. Overexpression of BCMA activates NF-κB and JNK signaling pathways. A chromosomal aberration in BCMA is found in a form of T-cell acute lymphoblastic leukemia (T-ALL).

Construct Details: Expressed as the combination of a heavy chain (HC) containing VH from anti-human BCMA mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-human BCMA mAb and CL of human kappa light chain.  Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition.

    

Source: Mammalian cells stably expressing anti-BCMA heavy chain and light chain (in human IgG1,kappa format) and growing in chemical-defined media with no animal components or antibiotics. 

 

M.W.: Calculated molecular mass (kDa): 146 (49 kDa for a single HC, 24 kDa for a single LC); Estimated by SDS-PAGE under reducing condition(kDa): 50 kDa + 25 kDa.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.

 

 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
