Description

T-cell surface glycoprotein CD3 epsilon chain, also known as CD3e, T3e, TCRe and IMD18, is a single-pass type I transmembrane protein. CD3E contains 1 Ig-like (immunoglobulin-like) domain in the extracellular region and 1 cytoplasmic ITAM (immune tyrosine activation motif) domain. CD3e, together with CD3-gamma, CD3-delta and CD3-zeta, and the T-cell receptor (TCR) alpha/beta and gamma/delta heterodimers, forms the TCR-CD3 complex. This complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways leading to the activation of T cells. CD3e plays an essential role in T-cell development. In CD3e-deficient mice, all stages of T cell development and activation that are TCR-dependent were impaired, but not eliminated. Mutations in the CD3e gene are associated with immunodeficiency 18 (IMD18), an autosomal recessive primary immunodeficiency characterized by onset in infancy or early childhood. The CD3e gene has also been linked to a susceptibility to type I diabetes in women.

Construct Details: Expressed as the combination of a heavy chain (HC) containing VH from anti-human CD3e mAb and CH region of mouse IgG1 and a mouse kappa light chain (LC) encoding VL from anti-CD3e mAb.  Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition.

 

Source: Mammalian cells stably expressing anti-CD3e heavy chain and light chain (in mouse IgG1 format) and growing in chemical-defined media with no animal components.

Endotoxin Level: <0.1 EU per 1 μg of purified recombinant protein determined by the LAL method

 

Biological Activity: Recognizes T cell surface marker CD3e by flow cytometry, IHC staining and fluorescent microscopy. Has the same binding specificity as anti-CD3e OKT3 mAbs and recombinant anti-CD3e mAbs, human IgG1 (SKU#MAB0818), and rabbit IgG (SKU#MAB1897).

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
