The TGFβ (transforming growth factor β) superfamily proteins are pleiotropic cytokines that regulate a diverse range of cellular processes, including proliferation, differentiation, migration, adhesion and death.  The TGFβ superfamily elicits 4 signaling pathways: TGFβ, Bone Morphogenic Protein (BMP), Activin, and Nodal.  Each pathway signals through a heteromeric receptor complex composed of at least two type-I and two type-II transmembrane receptors containing cytoplasmic serine/threonine kinase domains. These receptors are distinguished by the presence of a glycine/serine-rich juxta-membrane domain found only in the type I receptors. Either type receptor may initially bind ligand, followed by the recruitment of an alternate-type receptor counterpart to form a signal-activating complex. The type I receptors are also referred to as ALKs (Activin receptor-like kinases). ACVR1B (Activin receptor type-1B), also known as ALK4, SKR2, ActR-IB, and ACVRLK4, is a type I receptor in the TGFβ receptor (TGFBR) family.  ACVR1B acts as the main type I receptor to transduce signals of activin and activin-like ligands. Activin binds either ACVR2A or ACVR2B type II receptor and then forms a complex with ACVR1B to activate AVCR1B through phosphorylation. The activated ACVR1B in turn initiates downstream events by phosphorylation of its effectors to regulate gene expression and affect cellular phenotype. Activin signaling can be modulated by the direct interaction of ACVR1B with cripto or inhibin binding protein.  ACVR1B plays an important role in embryonic cell fate and axis determination, cell proliferation and apoptosis, as well as tumorigenesis.

Construct Details: Full length human ACVR1B/ALK4 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
