Description

ASTN2 (Astrotactin-2) is a two-pass type I transmembrane protein that belongs to the astrotactin family.  ASTN2 contains 2 extracellular domains (ECD), including a 157 aa (amino acid) long small ECD1 and a 884 aa long large ECD2.  The ECD2 is composed of three EGF-like motifs and one Fibronectin type-III motif.  The C-terminal region after the fibronectin type-III domain presents structural similarity to annexin domains and binds calcium ions. ASTN2 is expressed in the brain and may function in neuronal migration, based on functional studies of the related ASTN1.  ASTN2 mediates recycling of the neuronal cell adhesion molecule ASTN1 to the anterior pole of the cell membrane in migrating neurons. ASTN2 also promotes ASTN1 internalization and intracellular transport of endocytosed ASTN1. ASTN2 binds inositol-4,5-bisphosphate, inositol-3,4,5-trisphosphate and inositol-1,3,4,5-tetrakisphosphate, suggesting it is recruited to membranes lipids with a phosphoinositide headgroup. A deletion at the ASTN2 locus has been associated with schizophrenia.
