Description

The mannose 6-phophate receptors (MPRs) are single-pass type I transmembrane glycoproteins that target the newly synthesized hydrolases from the Golgi complex and the cell surface to lysosomes. There are two different MPRs, one of ~300kDa termed CI-MPR (cation-independent) and a smaller, dimeric receptor of ~46kDa termed CD-MPR (cation-depnendent). Both MPRss are P-type lectins that bind terminal mannose 6-phosphate (M6P) with similar affinity (CI-MPR = 7 μM, CD-MPR = 8 μM) and contain similar cytoplasmic signals for intracellular trafficking. CD-MPR shows greatly enhanced binding to M6P in the presence of divalent cations, such as manganese. The MPRs, when bound to their cargo via M6P, are recognized by the GGA family of clathrin adaptor proteins and accumulate in forming clathrin-coated vesicles. The vesicles are trafficked to the early endosome where, in the relatively low pH environment of the endosome, the MPRs release their cargo. The cargo proteins are then trafficked to the lysosome via the late endosome independently of the MPRs. CD-MPR is also known as M6PR that contain a 157 amino-acid M6P-binding-domain in the extracellular/luminal region. CI-MPR was also identified as the cell surface receptor for insulin-like growth factor 2 (IGF2R/CD222). IGF2R consists of 15 repeats homologous to the M6P-binding domain present in M6PR/CD-MPR and two of these 15 repeats are responsible for binding to M6P.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
