Description

NCR1, also known as NKp46 and CD335, is a single-pass type I transmembrane glycoprotein that contains two extracellular Immunoglobulin (Ig)-like domains and a short cytoplasmic tail.  CD335 together with NCR2/NKp44/CD336 and NCR3/NKp30/CD337 are activating receptors collectively termed the natural cytotoxicity receptors (NCRs). All these receptors lack significant sequence homology to one another. They are expressed almost exclusively by NK cells and are primarily responsible for triggering NK cell-mediated lytic activities of human tumor cells. CD335 and its co-receptor CD59 together activate cytotoxicity of human NK-cells by the association with ITAM bearing signal adapter proteins, including CD3ζ and FcεR1γ, which are able to form disulfidelinked homodimers and heterodimers. Blocking any of these three NCRs individually resulted in partial inhibition of tumor cell lysis, but nearly complete inhibition of lysis was observed if all three receptors were blocked simultaneously. CD335 is also implicated in NK cell-mediated lysis of pathogen-infected cell lines and mononuclear phagocytes infected with an intracellular bacterium. CD335 recognizes virusinfected cells through its capacity to bind to viral hemagglutinins.  CD335 is involved in NK-cell cell death by apoptosis.
