Description

CD47, also known as IAP and OA3, is a variably glycosylated transmembrane protein of the immunoglobulin superfamily (IgSF). It contains an extracellular domain (ECD) with a single Ig-like domain, five transmembrane domains with short intervening loops and a 34 amino acid cytoplasmic tail at the C-terminus. CD47 is broadly expressed. It is involved in the increase in intracellular calcium concentration in response to cell adhesion to extracellular matrix. By acting as an adhesion receptor, CD47 plays a role in both cell adhesion by positively or negatively modulating Integrin-mediated function. CD47 also binds to the SIRP family members on macrophages, neutrophils, and T lymphocytes, preventing macrophagemediated clearance of CD47expressing cells and promoting immune cell transmigration across the vascular endothelium.  CD47 binds SIRPA which prevents maturation of dendritic cells and inhibits cytokine production. CD47 also interacts with SIRPG that enhances superantigen-dependent T-cell-mediated proliferation and co-stimulates T-cell activation. CD47 is also a receptor for thrombospondin, and inhibits the angiogenic and vasorelaxant effects in the vasculature. On dendritic cells and myeloma cells, CD47 binds TSP-1 and induces giant cell formation and osteoclast differentiation.
