Description

MSR1 (macrophage scavenger receptor 1), also known as CD204 and SCARA1 (scavenger receptor class A member 1), is a single-pass, type II membrane protein.  It belongs to an expanding family of transmembrane molecules collectively referred to as scavenger receptors.  Receptors of this family bind to a series of negativelycharged/polyanionic ligands.  They recognize and internalize modified low density lipoproteins (LDLs) in the removal of many foreign substances and waste materials in the living body. Scavenger receptors can be categorized into 3 classes, and MSR1 is the class A receptors with 3 different types generated by alternative splicing. The extracellular region of MSR1 contains an αhelical coiledcoil domain, a collagenlike domain, and a cysteinerich domain (CRD) at the C-terminus. MSR1 is a macrophage-specific trimeric transmembrane glycoprotein involved in many macrophage-associated physiological and pathological processes including atherosclerosis, Alzheimer's disease, and host defense. The isoforms type 1 (MSR-AI or SR-AI) and type 2 (MSR-AII or SR-AII) are functional receptors, which mediate the endocytosis of modified LDLs. The isoform type 3 (MSR-AIII or SR-AIII) does not internalize modified LDLs. It has an altered intracellular processing and is trapped within the endoplasmic reticulum.  Thus the isoform type 3 can inhibit the function of isoforms type 1 and type 2 when co-expressed, indicating a dominant negative effect. MSR1 may also play important roles in macrophage growth, cell adhesion, and osteoclast differentiation.
