GPNMB (transmembrane glycoprotein NMB), also known as Osteoactivin and HGFIN (hematopoietic growth factor-inducible neurokinin 1 ), is a single pass type I transmembrane glycoprotein of the NMB/pMEL­17 family. GPNMB is expressed in many cell types, including osteoclasts, macrophages, fibroblasts, dendritic cells, retinal pigment epithelial cells, melanocytes and tumor cells. It is often associated with the endosomal/lysosomal compartment.  GPNMB is composed of a 465 aa luminal/extracellular domain (ECD), a transmembrane segment and a 53 aa cytoplasmic tail. The ECD contains a heparin­binding motif, multiple glycosylation sites, an RGD motif and an 88 aa PKD domain.  The cytoplasmic tail has an ITAM (Y­x­x­I) and lysosomal targeting (L­L) motif. GPNMB may paly a role in osteoblast maturation, cell adhesion and migration. GPNMB may mediate BMP-2 effects on osteoblast differentiation and function. GPNMB may also participates in bone mineralization, and acts as a negative regulator of inflammation in macrophages. GPNMB expression downregulates the macrophage inflammatory response by inhibiting IL­6 and IL­12 production.  GPNMB is reported to bind to heparan sulfate­proteoglycan and integrins in mice. GPNMB is expressed at high levels in normal and inflammatory liver macrophages implicating a role in acute liver injury. GPNMB may also play a role in triggering renal interstitial fibrosis through activation of matrix metalloproteinase expression and collagen remodeling. GPNMB is expressed in human breast cancers and is capable of promoting breast cancer.

Storage

 

The product is shipped at 4°C for immediate use. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
