Description

AGRL1 is a 7 transmembrane (7TM) receptor belonging to the secretin receptor family, which is also known as family B or 2 of the G-protein-coupled receptors (GPCRs). Many members of this family are receptors for peptide hormones from the glucagon hormone family and regulate the activation of two major signal transduction pathways: adenylyl cyclase (cAMP) and phosphatidylinositol-calcium pathways. AGRL1 is an orphan GPCR and may be proteolytically processed to form a heterodimer of 2 chains: the large extracellular N-terminal fragment and the membrane-bound C-terminal 7TM fragment predominantly remain associated and non-covalently linked. It forms a heterodimer, consisting of a large extracellular region (p120) non-covalently linked to a seven-transmembrane moiety (p85). AGRL1 interacts with syntaxin and with proteins of the SHANK family via the PDZ domain and with FLRT1, FLRT2 and FLRT3 via extracellular domain.  AGRL1 is a calcium-independent receptor of high affinity for alpha-latrotoxin, an excitatory neurotoxin present in black widow spider venom which triggers massive exocytosis from neurons and neuroendocrine cells. AGRL1 is also a receptor for TENM2 that mediates heterophilic synaptic cell-cell contact and postsynaptic specialization. AGRL1 has been implicated in the regulation of exocytosis.

Construct Details: Full length human AGRL1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
