Description

IL13RA2 (Interleukin-13 receptor subunit alpha-2), also known as CD213a2 and IL13BP (IL13-binding protein), is a single-pass type I transmembrane protein belonging to the type I cytokine receptor family. Like IL13RA1, the extracellular domain (ECD) of IL13RA1 contains 3 fibronectin type III domains, D1, D2, and D3, of which the last two comprise the cytokine receptor homology modules (CRHs).  In contrast to IL13RA1, IL13RA2 binds IL-13 with a high affinity but lacks cytoplasmic domain, and does not appear to function as a signal mediator. IL-13RA2 is widely expressed and detected on many types of cancer cells including glioblastoma, AIDS Kaposi's sarcoma, and head & neck cancer. IL-13 binds two types of receptor: (1) a heterodimer of the IL13 RA1 and the IL4RA, transducing the IL13-dependent proliferative signal; and (2) the IL13RA2, acting as a nonsignaling "decoy" receptor by binding and internalizing IL13. IL13RA2 can also block IL4-induced responses by inhibiting IL4 bound IL13RA1/IL4RA receptor complexes even though it does not bind IL 4 directly. Aside from its decoy function, IL13-IL13RA2 binding directly promotes the development of tissue fibrosis. IL13RA2 may also play an important role in the Th2 polarized immune responses characteristic of a variety of pathologies, including parasitic infections and allergic asthma. IL-13Rα2 exists both on the cell membrane and in soluble form which are generated from distinct transcripts.
