Description

CSF1 (colony stimulating factor 1), also known as M­CSF (macrophage colony stimulating factor) is a single-pass type I transmembrane or secreted protein. The active form of CSF1 is a disulfide-linked homodimer, produced by proteolytic cleavage of membrane-bound precursors.  As a cytokine, CSF1 controls the production, differentiation, and function of macrophages. CSF1 primes and enhances macrophage killing of tumor cells and microorganisms, regulates the release of cytokines and other inflammatory modulators from macrophages, and stimulates pinocytosis. CSF1 induces the release of proinflammatory chemokines, and thereby plays an important role in innate immunity and in inflammatory processes. It also plays an important role in the regulation of osteoclast proliferation and differentiation, the regulation of bone resorption, and is required for normal bone development. CSF1 is required for male and female fertility. CSF1 induces reorganization of the actin cytoskeleton, regulates formation of membrane ruffles, cell adhesion and cell migration. Aberrant expression of CSF1 or its receptor CSF1R can promote cancer cell proliferation, invasion and formation of metastases. Overexpression of CSF1 or CSF1R is observed in many types of cancers including breast, ovarian, prostate, and endometrial cancers.  Aberrant expression of CSF1 or CSF1R is implicated in inflammatory conditions, such as rheumatoid arthritis (RA), glomerulonephritis, atherosclerosis, and allograft rejection.

Construct Details: Full length human CSF1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
