Description

PLXNB2 (Plexin B2) is a single-pass type I transmembrane protein belonging to the B class of plexins.  PLXNB2 contains 1 SEMA and 3 IPT/TIG domains in its large extracellular region. PLXNB2 exists as a monomer or heterodimer with PLXNB1.  PLXNB2 is a cell surface receptor for SEMA4C, SEMA4D and SEMA4G, which are class 4 semaphorins with important roles in cell-cell signaling. Binding to various semaphorins promotes downstream activation of RHOA and phosphorylation of ERBB2/HER2. PLXNB2 also interacts with MET, ARHGEF11/12, and MST1R. PLXNB2 is required for normal differentiation and migration of neuronal cells during brain corticogenesis and for normal embryonic brain development. PLXNB2 regulates the migration of cerebellar granule cells in the developing brain. It plays a role in RHOA activation and subsequent changes of the actin cytoskeleton as well as in axon guidance, invasive growth and cell migration. PLXNB2 may modulate the activity of RAC1 and CDC42.

Construct Details: Full length human PLXNB2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

 

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Primers: Gene insert coding sequence can be confirmed by following primers:

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
