LILRB4 (Leukocyte immunoglobulin like receptor B4), also known as CD85k, LIR5 (Leukocyte immunoglobulin-like receptor 5), and ILT3 (Ig-like transcript5), is a member of the leukocyte immunoglobulin (Ig)-like receptor (LIR) family of the Ig superfamily (IgSF).  It belongs to the subfamily B class of LIR receptors (LILRBs), which are single-pass type I trnasmembrane proteins with 2 or 4 extracellular Ig-like domains, a transmembrane domain (TM), and 2-4 cytoplasmic immunoreceptor tyrosine-based inhibitory motifs (ITIMs). LILRB1 contains 2 Ig-like C2 type domains and 3 TIIMs. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases to modulate cellular responses.  LILRB1 is detected in monocytes, macrophages, dendritic cells, lung, natural killer cells and B-cells. LILRB4 acts as a receptor for class I MHC antigens. It recognizes a broad spectrum of HLA-A, HLA-B, HLA-C and HLA-G alleles. LILRB4 may be involved in the down-regulation of the immune response and the development of tolerance, e.g. towards transplants. LILRB4 interferes with TNFRSF5-signaling and NF-kappa-B up-regulation. It also inhibits receptor-mediated phosphorylation of cellular proteins and mobilization of intracellular calcium ions.

Construct Details: Full length human LILRB4 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells.  The construct also contains a Puromycin marker for antibiotic stable selection. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
