Description

CD125, also known as IL5RA/IL5R (Interleukin-5 receptor subunit alpha), is a single pass, type I transmembrane glycoprotein of the type I cytokine receptor family, type 5 subfamily.  CD125/IL5RA consists of an extracellular region with 2 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, a transmembrane domain, and a cytoplasmic tail with the box 1 motif.  The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction/activation. CD125/IL5RA is expressed on eosinophils and basophils. CD125/IL5RA is a component of the receptor complex specific for interleukin-5. The IL5 receptor complex is a heterodimer of CD125/IL5RA and the common signal transducing subunit CD131/CSF2RB shared by the receptors for interleukin 3 (IL3), colony stimulating factor 2 (CSF2/GM-CSF), and interleukin 5 (IL5). The binding of CD125 to IL5 depends on the beta subunit CD131. The beta subunit CD131 is activated by the ligand binding, and is required for the biological activities of IL5. This protein has been found to interact with syndecan binding protein (syntenin), which is required for IL5 mediated activation of the transcription factor SOX4.

Construct Details: Full length human IL5RA/CD125 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
