Description

HFE (hereditary hemochromatosis protein) is a single-pass type I transmembrane protein belonging to the Immunoglobulin superfamily (IgSF).  HFE contains an Ig-like C1-type domain in the extracellular region.  HFE is similar to MHC class I-type proteins and associates with beta2-microglobulin (beta2M). It is thought that HFE functions to regulate iron absorption by regulating the interaction of the transferrin receptor (TfR) with transferrin. The iron storage disorder, hereditary haemochromatosis, is a recessive genetic disorder that results from defects in the HFE gene. HFE binds TfR through the extracellular domain in a pH-dependent manner.  Variations affecting the HFE gene are also associated with Variegate porphyria (VP) and  Microvascular complications of diabetes 7 (MVCD7)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
