Description

KLRB1 belongs to the killer cell lectin-like receptor (KLR) or NKG2 family of the C-type lectin superfamily, which is a group of single pass type II transmembrane proteins preferentially expressed in natural killer (NK) cells.  NK cells are lymphocytes that can mediate lysis of certain tumor cells and virus-infected cells without previous activation. NK cells regulate specific humoral and cell-mediated immunity. The KLR/NKG2 family is characterized by the presence of a C-type lectin domain in the extracellular region at the C-terminus. KLRK1 forms a disulfide-bond linked  homodimer. Activation of KLRB1 results in specific acid sphingomyelinase/SMPD1 stimulation with subsequent marked elevation of intracellular ceramide. It also leads to AKT1/PKB and RPS6KA1/RSK1 kinases stimulation as well as markedly enhanced T-cell proliferation induced by anti-CD3.  KLRB1 acts as a lectin that binds to the terminal carbohydrate Gal-alpha(1,3)Gal epitope as well as to the N-acetyllactosamine epitope. It also bindo to CLEC2A and CLEC2D as a ligand and inhibits NK cell-mediated cytotoxicity as well as interferon-gamma secretion in target cells.

Construct Details: Full length human KLRB1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
