Description

CD207 or CLEC4K (C-type lectin domain family 4 member K) is a single-pass type II transmembrane protein and a member of the C-type lectin superfamily.  CLEC4K/CD207 contains 1 C-type lectin domain in the extracellular region. CLEC4K is expressed only in Langerhans cells which are immature dendritic cells of the epidermis and mucosa. CLEC4K is localized in the Birbeck granules, organelles present in the cytoplasm of Langerhans cells and consisting of superimposed and zippered membranes. CLEC4K is a C-type lectin with mannose binding specificity, and it has been proposed that mannose binding by this protein leads to internalization of antigen into Birbeck granules and providing access to a nonclassical antigen-processing pathway. Mutations in the CLEC4K gene result in Birbeck granules deficiency or loss of sugar binding activity. CLEC4K binds to sulfated as well as mannosylated glycans, keratan sulfate (KS) and beta-glucans. It facilitates uptake of antigens and is involved in the routing and/or processing of antigen for presentation to T cells. CLEC4K acts as a major receptor on primary Langerhans cells for Candida species, Saccharomyces species, and Malassezia furfur. CLEC4K protects against human immunodeficiency virus-1 (HIV-1) infection. CLEC4K binds to high-mannose structures present on the envelope glycoprotein followed by subsequent targeting of the virus to the Birbeck granules leading to its rapid degradation.

Construct Details: Full length human CD207 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
