Description

MRC2 (mannose receptor C type 2), also known as CD280, UPARAP (Urokinase-type plasminogen activator receptor-associated protein) and CLEC13E (C-type lectin domain family 13 member E), is a single-pass type I transmembrane protein and a member of the mannose receptor family of the C-type lectin superfamily.  MRC2 contains  8 C-type lectin domains, 1 fibronectin type-II domain and 1 ricin B-type lectin domain in the extracellular region.  MRC2 plays a role in extracellular matrix remodeling by mediating the internalization and lysosomal degradation of collagen ligands. MRC2 m play a role as endocytotic lectin receptor displaying calcium-dependent lectin activity. It internalizes glycosylated ligands from the extracellular space for release in an endosomal compartment via clathrin-mediated endocytosis. MRC2 may be involved in plasminogen activation system controlling the extracellular level of PLAUR/PLAU, and thus may regulate protease activity at the cell surface. MRC2 may contribute to cellular uptake, remodeling and degradation of extracellular collagen matrices. It may play a role during cancer progression as well as in other chronic tissue destructive diseases acting on collagen turnover. MRC2 may participate in remodeling of extracellular matrix cooperating with the matrix metalloproteinases (MMPs).  Expression of MRC2 may play a role in the tumorigenesis and metastasis of several malignancies including breast cancer, gliomas and metastatic bone disease.

Construct Details: Full length human CD280 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Transduction Protocol

 

The product is shipped at 4°C for immediate use.  Upon receipt, centrifuge the vial briefly before opening.  Store at –80°C or lower and the product is stable for 3 months.  Avoid repeated freeze-thaw cycles.

 

 

The product should be employed in a Biosafety Level 2 tissue culture facility.
