Description

CD126, also known as IL6RA (Interleukin-6 receptor subunit alpha), is a single pass, type I transmembrane glycoprotein of the type I cytokine receptor family, type 3 subfamily.  IL6RA/CD126 consists of an extracellular region with 1 Ig-like C2-type (immunoglobulin-like) and 2 fibronectin type III (FN3) domains, in addition to a transmembrane (TM) domain, and a cytoplasmic tail. IL6 is a potent pleiotropic cytokine that regulates cell growth and differentiation and plays an important role in the immune response. IL6RA/CD126 itself binds to IL6 with low affinity and does not transduce a signal. IL6RA/CD126 is a subunit of the IL6 receptor complex (IL6R) consisting of CD126 and interleukin 6 signal transducer (IL6ST/GP130/IL6-beta), a receptor subunit also shared by many other cytokines. Signal activation requires IL6ST and activation may lead to the regulation of the immune response, acute-phase reactions and hematopoiesis. Dysregulated production of IL6 and its receptor IL6RA are implicated in the pathogenesis of many diseases, such as multiple myeloma, autoimmune diseases and prostate cancer. Low concentration of a soluble form of IL6RA may act as an agonist of IL6 activity.

Construct Details: Full length human IL6RA/CD126 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
