Description

CDC1P1 (CUB domain-containing protein 1) or CD318 is an 836 amino acid (aa) single pass type I transmembrane protein. CDCP1 contains an N-terminal signal peptide, a 638-aa extracellular domain (ECD) with 3 CUB domains and 7 potential glycosylation sites, a transmembrane domain (TM), and a 148-aa cytoplasmic tail. DCP1 is highly expressed in mitotic cells with low expression during interphase. It is expressed in cells with phenotypes reminiscent of mesenchymal stem cells and neural stem cells. CDCP1 is up-regulated in a number of human tumor cell lines, as well as in colorectal cancer, breast carcinoma and lung cancer. CDCP1 interacts with CDH2/N-cadherin, CDH3/P-cadherin, SDC1/syndecan-1, SDC4/syndecan-4 and the serine protease ST14/MT-SP1.  CDCP1 acts as a substrate for Src family kinases such as SRC and YES as well as by the protein kinase C gamma/PRKCG.  CDCP1 may be involved in cell adhesion and cell matrix association. CDCP1 may play a role in the regulation of anchorage versus migration or proliferation versus differentiation via its phosphorylation.  CDCP1 belongs to the tetraspanin web involved in the tyrosine phosphorylation-dependent regulation of tumor invasion and metastasis. CDCP1 may be a novel marker for leukemia diagnosis and for immature hematopoietic stem cell subsets.  A soluble form of CDCP1 can be produced by proteolytic cleavage at an unidentified site on its N-terminal side and converted to p80 by plasmin, a trypsin-like protease.

Construct Details: Full length human CDCP1/CD3218 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
