Description

Cadherins are calcium-dependent cell adhesion proteins that preferentially interact with themselves in a homophilic manner in connecting cells.  Cadherins thus contribute to the sorting of heterogeneous cell types.  Cadherin4 or CDH4 is a 916 amino acid (aa) single pass type I transmembrane protein of the cadherin superfamily.  CDH4 contains an N-terminal signal peptide, a 565-aa extracellular domain (ECD) with 5 cadherin domains and 6 potential glycosylation sites, a transmembrane domain (TM), and a 160-aa highly conserved cytoplasmic tail.  CDH4 may play an important role during brain segmentation and neuronal outgrowth as well as in kidney and muscle development. Of particular interest are studies showing stable cis-heterodimers of cadherins 2 and 4 in co-transfected cell lines. Previously thought to interact in an exclusively homophilic manner, this is the first evidence of cadherin heterodimerization.

Construct Details: Full length human CDH4 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

 

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
