Description

GFP (green fluorescent protein) was discovered in Aequorea victoria (Jellyfish).  GFP transduces the blue chemiluminescence of aequorin into green fluorescent light upon receiving energy from the Ca2+-activated photoprotein aequorin. Its chromophore is formed by autocatalytic backbone condensation between Ser-65 and Gly-67, and oxidation of Tyr-66 to didehydrotyrosine. GFP has been engineered to produce a large number of differently colored variants for making chimeric proteins as a fluorescent protein tag. GFP proteins can be expressed in most known cell types as a noninvasive marker in living cells and organisms. Therefore GFP proteins enable a wide range of applications where they can act as a cell lineage tracer, reporter of gene expression, as a measure of protein-protein interactions, or as a molecular biosensor and thermometer.

Construct Details: GFP (encoding UniProt #P42212) is cloned into the Lentiviral expression vector pLTC with an EF1α promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It was co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of antibiotic stable-selection markers are available.

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl inα sterile TE buffer (pH8.0)

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

 

 

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
