Description

JAM3 (Junctional adhesion molecule 3) orJAMC is a 310 amino acid (aa) single pass type I transmembrane glycoprotein that belongs to the Immunoglobulin superfamily (IgSF).  JAM3 contains an N-terminal signal peptide, a 210-aa extracellular domain (ECD) with 2 Ig-like V-type domains and 3 potential glycosylation sites, a transmembrane domain (TM), and a 48-aa short cytoplasmic tail. JAM3 is localized in the tight junctions between high endothelial cells. Tight junctions represent one mode of cell-to-cell adhesion in epithelial or endothelial cell sheets, forming continuous seals around cells and serving as a physical barrier to prevent solutes and water from passing freely through the paracellular space. JAM3 acts as a receptor for another member of this family JAM2, which togther form a heterodimer.  JAM3 participates in cell-cell adhesion as a counter-receptor for ITGAM, mediating leukocyte-platelet interactions and is involved in the regulation of transepithelial migration of polymorphonuclear neutrophils (PMN). The soluble form of JAM3 is thought to be a mediator of angiogenesis. A mutation in an intron of the JAM3 gene is associated with hemorrhagic destruction of the brain, subependymal calcification, and congenital cataracts (HDBSCC).

Construct Details: Full length human JAM3 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
