Description

OLR1 (oxidized low-density lipoprotein receptor 1) or LOX1 (lectin-like oxidized LDL receptor 1) is a single-pass type II transmembrane protein that belongs to a the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily. OLR1 contains 1 coiled coil, 1 tandem-repeat neck domain and 1 C-type lectin domain in the extracellular region. OLR1 acts as  a LDL receptor and is regulated through the cyclic AMP signaling pathway. OLR1 binds, internalizes and degrades oxidized low-density lipoprotein (oxLDL). oxLDL is a marker of atherosclerosis that induces vascular endothelial cell activation and dysfunction, resulting in pro-inflammatory responses, pro-oxidative conditions and apoptosis.  OLR1 may be involved in the regulation of Fas-induced apoptosis and play a role as a scavenger receptor.  OLR1 also acts as a receptor for the HSP70 protein involved in antigen cross-presentation to naive T-cells in dendritic cells, thereby participating in cell-mediated antigen cross-presentation. OLR1 is involved in inflammatory process, by acting as a leukocyte-adhesion molecule at the vascular interface in endotoxin-induced inflammation. OLR1 is also  a receptor for advanced glycation end (AGE) products, activated platelets, monocytes, apoptotic cells and both Gram-negative and Gram-positive bacteria. Mutations of the OLR1 gene have been associated with atherosclerosis, risk of myocardial infarction, and may modify the risk of Alzheimer's disease.

Construct Details: Full length human OLR1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
