Description

NTRK1 (neurotrophic receptor tyrosine kinase 1) receptor tyrosine kinase (RTK), is a single-pass type I transmembrane glycoprotein that belongs to the insulin receptor (InsR) subfamily of the tyrosine protein kinase family. NTRK1 comprises a 391-aa extracellular domain (ECD), a transmembrane region (TM), and a 357-aa intracellular kinase domain. The ECD contains 2 LRR (leucine-rich repeat) domains, 1 LRRCT and 2 Ig-like C2-type domains.  NTRK1 is involved in the development and the maturation of the central and peripheral nervous systems through regulation of proliferation, differentiation and survival of sympathetic and nervous neurons. NTRK1 acts as a high affinity receptor for NGF which is its primary ligand. NTRK1 can also bind and be activated by NTF3/neurotrophin-3. However, NTF3 only supports axonal extension through NTRK1 but has no effect on neuron survival.  Upon neurotrophin binding, NTRK1 phosphorylates itself and members of the MAPK pathway. In absence of ligand and activation, NTRK1 may promote cell death, making the survival of neurons dependent on trophic factors.  Mutations in the NTRK1 gene have been associated with congenital insensitivity to pain, anhidrosis, self-mutilating behavior, mental retardation and cancer.
