Description

Vascular endothelial cell growth factors (VEGFs) are secreted polypeptides with a highly conserved cystine-knot structure similar to that of the platelet-derived growth factors (PDGFs).  VEGFs play important roles in vascular development and in diseases involving abnormal growth of blood vessels such as tumor-related angiogenesis. VEGFs stimulate angiogenesis by binding to the specific receptors (VEGFRs) on the surface of vascular endothelial cells. All the three VEGFRs, VEGFR1 (Flt-1), VEGFR2 (KDR/Flk-1/CD309), and VEGFR3 (Flt-4), belong to the CSF-1/PDGF receptor subfamily of receptor tyrosine kinase superfamily (RTKs). Each VEGFR contains 7 immunoglobulin (Ig)-like C2-type domains in the extracellular region and an intracellular kinase domain. VEGFR1 is a receptor for VEGF, VEGFB and PDGF. VEGFR1 is expressed predominantly in normal lung and specifically expressed in most of the vascular endothelial cells and peripheral blood monocytes. VEGFR1 plays an essential role in the development of embryonic vasculature, the regulation of angiogenesis, cell survival, cell migration, macrophage function, chemotaxis, and cancer cell invasion. It may act as a negative regulator of embryonic angiogenesis by inhibiting excessive proliferation of endothelial cells. VEGFR1 can promote endothelial cell proliferation, survival and angiogenesis in adulthood. VEGFR1 shows very high affinity for VEGF and relatively weak protein kinase activity.  VEGFR1 may function as a negative regulator of VEGF signaling by limiting the amount of free VEGF and preventing the binding to VEGFR2/KDR.

Construct Details: Full length humanFLT1 gene (encoding Uniprot accession # P17948 - isoform 2 or soluble FLT1) is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
