Description

EPOR (erythropoietin receptor) is a single pass type I transmembrane glycoprotein of the type I cytokine receptor family (type 1 subfamily). EPOR is the erythropoietin receptor. Upon ligand binding, EPOR activates JAK2 (Janus kinase) tyrosine kinase which in turn activates different intracellular pathways including: Ras/MAP kinase (MAPK), phosphatidylinositol 3-kinase (PI3K) and STAT transcription factors.  EPOR consists of a 226 amino acid (aa) extracellular region with 1 fibronectin type III (FN3) domain and a juxtamembrane WSXWS motif, a transmembrane domain, and a 235 aa cytoplasmic region with a Box 1 motif and an ITIM (immunoreceptor tyrosine-based inhibitor motif). The WSXWS motif may be necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding.  The box 1 motif is required for JAK interaction/activation while the ITIM is involved in modulation of cellular responses. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases.that mediates the activation of JAK- Stat and MAPK as well as PI3K pathways.  EPOR plays an important role in erythroid cell proliferation and differentiation. Defects in the EPOR gene may produce erythroleukemia and familial erythrocytosis. Dysregulation of the EPOR gene may affect the growth of certain tumors.

Construct Details: Full length human EPOR gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
