Description

CXADR (coxsackievirus and adenovirus receptor) is a single-pass type I transmembrane protein belonging to the immunoglobulin (Ig) superfamily (IgSF).  It contains two Ig-like C2-type domains in the extracellular region (ECD) and a PDZ-binding motif in the cytoplasmic domain (ICD).  The Ig-like C2-type 1 domain mediates homodimerization and interaction with JAML, a transmembrane protein of the plasma membrane of leukocytes. The PDZ-binding motif mediates interaction with signaling molecules such as MPDZ and BAIAP1.  CXADR acts as a receptor for group B coxsackieviruses and subgroup C adenoviruses.  CXADR is a component of the epithelial apical junction complex that may function as a homophilic cell adhesion molecule and is essential for tight junction integrity. CXADR is also involved in transepithelial migration of leukocytes through adhesive interactions with JAML. The interaction between these two receptors also mediates the activation of gamma-delta T-cells, a subpopulation of T-cells residing in epithelia and involved in tissue homeostasis and repair. Upon epithelial CXADR-binding, JAML induces downstream cell signaling events in gamma-delta T-cells through PI3-kinase and MAP kinases. This results in proliferation and production of cytokines and growth factors by T-cells that in turn stimulate epithelial tissues repair.

Product Details

Gene Symbol: CXADR; CAR; HCAR; CAR4/6; HCVADR

NCBI Gene ID: 1525

Uniprot Entry: P78310

Construct Details: Full length human CXADR gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Gene Insert Size: 1098 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
