Description

CD177 is a glycosyl-phosphatidylinositol (GPI)-anchored cell surface glycoprotein that plays a role in neutrophil activation. A variety of GPI-linked surface glycoproteins are composed of one or more copies of a conserved domain of ~100 amino-acid residues termed UPAR/Ly6 motif that was originally found in UPAR (Urokinase plasminogen activator surface receptor) and Ly6/CD59. The u-PAR/Ly-6 (UL) motif has 10 conserved cysteines with a defined disulfide-bonding pattern.  CD177 contains 2 copies of  UPAR/Ly6 motifs in the extracellular domain (ECD).  CD177 is highly expressed in normal bone marrow and weakly expressed in fetal liver. CD177 is detected  on neutrophils and in granulocytes of patients with polycythemia vera (PV) and with essential thrombocythemia (ET). A soluble form of CD177 may be produced by proteolytic cleavage at the cell surface (shedding).  CD177 binds platelet endothelial cell adhesion molecule-1 and function in neutrophil transmigration. Mutations in the CD177 gene are associated with myeloproliferative diseases. Over-expression of the CD177 gene has been found in patients with polycythemia rubra vera. Autoantibodies against CD177 may result in pulmonary transfusion reactions, and CD177 may be involved in Wegener's granulomatosis.

Construct Details: Full length human CD177 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
