Description

CD200R1 (Cell surface glycoprotein CD200 receptor 1) is a single pass type I transmembrane glycoprotein belonging to the CD200R family of the immunoglobulin (Ig) superfamily (IgSF). Like CD200, CD200R1 contains 2 extracellular Ig-like V-type (IgV) and Ig-like C2-type (IgC2) domains, a transmembrane and a cytoplasmic domain. CD200R1 is a receptor for CD200 and they interact via their respective N-terminal Ig-like domains.  CD200R1 is restricted to the surfaces of myeloid lineage cells.  It is primarily expressed in granulocytes, monocytes, most T-cells, neutrophils, basophils and a subset of NK, NKT and B-cells.  CD200R1 acts as an inhibitory receptor for the CD200/OX2 cell surface glycoprotein. The CD200:CD200R1 interaction may function as a myeloid down regulatory signal and this interaction may control myeloid function in a tissue-specific manner. CD200R1 limits inflammation by inhibiting the expression of proinflammatory molecules including TNF-alpha, interferons, and inducible nitric oxide synthase (iNOS) in response to selected stimuli. CD200R1 also binds to Human herpesvirus 8 (HHV-8) K14 viral CD200 homolog with identical affinity and kinetics as the host CD200.

Uniprot Entry: Q8TD46

Construct Details: Full length human CD200R1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
