Description

CXCR2 (C-X-C chemokine receptor type 2) is a 7-transmembrane protein of the G protein-coupled receptor 1 (GPCR1) family. CXCR2 acts as a receptor to interleukin-8 (IL8), which is a powerful neutrophils chemotactic factor. Binding of IL-8 to the receptor causes activation of neutrophils. This response is mediated via a G-protein that activate a phosphatidylinositol-calcium second messenger system. CXCR2 binds to IL-8 with high affinity and also binds with high affinity to CXCL3, CXCL1 and NAP-2. CXCL1/GRO/MGSA is a protein with melanoma growth stimulating activity, and has been shown to be a major component required for serum-dependent melanoma cell growth. CXCR2 mediates neutrophil migration to sites of inflammation. The angiogenic effects of IL8 in intestinal microvascular endothelial cells are found to be mediated by CXCR2. The CXCR2 gene, CXCR1/CD181/IL18RA, a gene encoding another high affinity IL8 receptor, as well as IL8RBP, a pseudogene of IL8RB, form a gene cluster in a region mapped to chromosome 2q33-q36. Knockout studies in mice suggested that CXCR2 controls the positioning of oligodendrocyte precursors in developing spinal cord by arresting their migration.

Construct Details: Full length human CXCR2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
