CD124, also known as IL4RA (Interleukin-4 receptor subunit alpha), is a single pass, type I transmembrane glycoprotein of the type I cytokine receptor family, type 4 subfamily.  CD124/IL4RA consists of an extracellular region with 1 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, a transmembrane domain, and a cytoplasmic tail with a box 1 motif and an immunoreceptor tyrosine-based inhibitor motif (ITIM). This ITIM is involved in modulation of cellular responses. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases.  The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction and activation. CD124/IL4RA acts as a receptor for both IL4 and IL13 to regulate IgE production. The IL4 response is involved in promoting Th2 differentiation. The IL4/IL13 responses are also involved in chemokine and mucus production at sites of allergic inflammation.  A soluble form of CD124 can be produced by proteolysis of the membrane-bound protein, and this soluble form can inhibit IL4-mediated cell proliferation and IL5 upregulation by T-cells. Allelic variations in the CD124 gene have been associated with atopy, a condition manifested by allergic rhinitis, sinusitus, asthma, or eczema. Polymorphisms in the CD124 gene are also associated with resistance to human immunodeficiency virus type-1 infection.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
