LGR5 (Leucine-rich repeat-containing G-protein coupled Receptor 5) is a member of the G-protein coupled receptor 1 (GPCR1) family of 7-pass transmembrane proteins. LGR5 contains a large N-terminal extracellular domain that contains leucine-rich repeats important for the formation of a horseshoe-shaped interaction motif for ligand binding.  LGR5 is the receptor for R-spondins (RSPO1, RSPO2, RSPO3 or RSPO4) that potentiates the canonical Wnt signaling pathway.  In contrast to classical G-protein coupled receptors, LGR5 does not activate heterotrimeric G-proteins to transduce the signal. Upon binding to R-spondins, LGR5 associates with phosphorylated LRP6 and frizzled receptors that are activated by extracellular Wnt receptors, triggering the canonical Wnt signaling pathway to increase expression of target genes. LGR5 is involved in the development and maintenance of the adult intestinal stem cells.  LGR5 is a stem cell marker of the intestinal epithelium and the hair follicle.

Construct Details: Full length human LGR5 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
