Description

UMOD (Uromodulin) or THP (Tamm-Horsfall urinary glycoprotein) is the most abundant protein in urine under physiological conditions.  UMOD is a glycosyl phosphatidylinositol (GPI)-anchored protein that is situated on the luminal cell surface of the loop of Henle.  UMOD can be secreted into urine following proteolytic cleavage of its extracellular region that contains 4 EGF-like motifs and a ZP domain that mediates polymerization, leading to the formation of long filaments.  UMOD functions in biogenesis of apical membrane of epithelial cells of the thick ascending limb of Henle's loop (TALH), where it promotes formation of complex filamentous gel-like structure to mediate the water barrier permeability. UMOD may also serve as a receptor for binding and endocytosis of cytokines (IL-1, IL-2) and TNF.  In the urine, UMOD may contribute to colloid osmotic pressure, retard passage of positively charged electrolytes, and act as a constitutive inhibitor of calcium crystallization.  Its excretion into urine provides defense against urinary tract infections.  Defects in the UMOD gene are associated with several renal disorders, including tubulointerstitial kidney disease, autosomal dominant, 1 (ADTKD1), medullary cystic kidney disease-2 (MCKD2), glomerulocystic kidney disease with hyperuricemia and isosthenuria (GCKDHI), and familial juvenile hyperuricemic nephropathy (FJHN).

Gene Symbol: UMOD; THP; Uromodulin; FJHN; HNFJ; THGP; HNFJ1; MCKD2; ADTKD1; ADMCKD2

NCBI Gene ID: 7369

Uniprot Entry: P07911

Construct Details: Full length human UMOD gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
