Human ALPP/PLAP/SEAP Protein, ECD (Extracellular Domain), Fc-fusion, Biotinylated, Recombinant
SEAP (secreted embryonic alkaline phosphatase) is a reporter widely used to study promoter activity or gene expression. SEAP is a truncated form of human placental alkaline phosphatase (PLAP/ALPP) by deletion of the GPI anchor. PLAP/ALPP is a metalloenzyme that catalyzes the hydrolysis of phosphoric acid monoesters. It belongs to a multigene family composed of four alkaline phosphatase isoenzymes. The enzyme functions as a homodimer and has a catalytic site containing one magnesium and two zinc ions, which are required for its enzymatic function. The protein is primarily expressed in placental and endometrial tissue. Strong ectopic expression has been detected in ovarian adenocarcinoma, serous cystadenocarcinoma, and other ovarian cancer cells. PLAP is extremely heat stable and resistant to the inhibitor such as L-homoarginine. SEAP is secreted into cell culture supernatant and offers many advantages over intracellular reporters. It reduces background alkaline phosphatase activity and facilitates the automation of the sampling and assay processes. It does not require the preparation of cell lysates and thus allows the continuous determination of reporter activity for kinetic analysis without disturbing the cells. There are many commercially available systems that may be employed to assay SEAP activity.
