Description

Terminally differentiated somatic cells can be reprogrammed to generate induced pluripotent stem (iPS) cells by overexpression of a defined set of transcription factors. Typically OCT4, SOX2, c-Myc and KLF4 (OKSM) are the 4 reprogramming factors that together are sufficient to reprogram differentiated cells to an embryonic-like state termed iPS (induced pluripotent stem) cells. The OKSM combination was identified initially by screening 24 pre-selected factors in mouse embryonic fibroblasts (MEFs). The iPS cells are morphologically and phenotypically similar to embryonic stem (ES) cells and offer exciting possibilities in stem cell research and regenerative medicine. This lentivector gene clone contains all these 4 human reprogramming transcription factors that are expressed from a single transcript using 2A peptide-mediated self-cleaving elements: Oct4-2A-Sox2-2A-Klf4-2A-c-Myc.

Construct Details: Full length mouse Oct4, Sox2, Klf4 and c-Myc genes linked by self-cleaving 2A-elements is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. 

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)

 

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
