Description

CD116, also known as GM-CSFRα (granulocyte macrophage colony stimulating factor receptor alpha), is a component of the receptor complex that mediates cellular responses to GM­CSF. This receptor complex is composed of a cytokine-specific alpha- chain CD116 and a beta-chain CD131 shared with the receptors for IL-3 and IL-5. CD116 alone binds GM-CSF with low affinity, whereas CD131 does not bind GM-CSF by itself. Association with CD131 converts CD116 from a low affinity to a high affinity receptor for GM­CSF. GM­CSF is a hematopoietic cytokine that regulates differentiation, survival, and proliferation of colony-forming unit-granulocyte-macrophage progenitor cells. It promotes the differentiation and mobilization of granulocyte­macrophage, erythroid, megakaryocyte, and eosinophil progenitors. GM­CSF also enhances the activation of myeloid cell effector functions and plays an important role in the development of Th1 biased immune responses, allergic inflammation, and autoimmunity. CD116 is a single pass, type I transmembrane glycoprotein that consists of a 298 amino acid (aa) extracellular region with 2 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, a transmembrane domain, and a 54 aa cytoplasmic tail that mediates the activation of Janus kinase (Jak)- Stat and MAPK as well as phosphatidylinositol 3-kinase (PI3K) pathways.  CD116 is expressed on hematopoietic stem cells, progenitor and differentiated cells in the myeloid lineage, vascular endothelial cells, and placenta as well as in those non­hematopoietic tumor cells.  Soluble forms of CD116 retain the ability to bind GM­CSF.

Construct Details: Full length human CD116 gene (encoding UniProt #P15509) is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
