Description

B-cell differentiation antigen CD72, also known as Lyb2, is a single pass type II transmembrane glycoprotein that plays an important role in adaptive immunity. CD72 contains a 95 amino acid (aa) cytoplasmic domain with 2 immunoreceptor tyrosine-based inhibitory motifs (ITIMs), a 21 aa transmembrane segment, and a 243 aa extracellular domain (ECD) with a coiled-coil domain and a C-type lectin domain. CD72 is expressed on B lineage cells including Pre-B-cells and B-cells but not terminally differentiated plasma cells, NK cells, monocytes, dendritic cells, and mast cells. CD5 exists as a disulfide-linked homodimer. It interacts with CD5 and with CD100/Semaphorin 4D.  CD5 also associates with CD79a in the B cell antigen receptor (BCR) complex following antigen stimulation.  CD5 inhibits BCR signaling through interactions with the phosphatase PTPN6/SHP-1. CD72 ligation with antibodies or with CD100 induces tyrosine dephosphorylation of the CD72 cytoplasmic domain and its dissociation from SHP-1, leading to B cell proliferation. CD100 is expressed on activated NK cells and the adhesive interaction between CD100+ NK cells and CD72+ cells leads to an enhanced killing and IFN­γ secretion.  Both CD72 and CD100 are required for the maintenance of B cell anergy and tolerance.  Mice that lack either CD72 or CD100 develop autoimmunity.  CD72 may also induce positive signaling in B cells independent of the BCR signaling.

Construct Details: Full length human CD72 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
